Sergey Korotkov researches the effects of heavy metals (Tl+, Cd2+), as well as Y3+, lanthanides (La3+, Pr3+, Nd3+, Gd3+) and some xenobiotics (diazoxide, pinacidil) on the properties of isolated rat and heart mitochondria. Oxygen consumption rate (respiration) is studied in experiments with energized mitochondria being in various energetic states of 4, 3, and 3UDNP (uncoupled 2,4-dinitrophenol, DNP). The ionic permeability of the inner mitochondrial membrane (IMM) is estimated by the swelling of mitochondria to the optical density change in different salt media. The inner membrane potential (ΔΨmito) is studied to change of the safranin fluorescence in a mitochondrial suspension. In addition, the mitochondrial permeability transition opening in the IMM is studied in the presence of Ca2+ and the above agents.
